Phytochemical fingerprint profiling of Rasapachaka Yoga and Katutumbi Taila using High-Performance Thin Layer Chromatography

Abstract
Background: Standardisation of Ayurveda formulations is essential to ensure their quality, safety, and therapeutic efficacy. Rasapachaka Yoga and Katutumbi Taila (medicated oil) are classical formulations described in Ayurveda for the management of metabolic disorders and glandular enlargement. Scientific validation of these formulations using modern analytical techniques is necessary to establish reliable quality control parameters. Objective: The present study is aimed at evaluating the organoleptic properties, physicochemical parameters, and high-performance thin-layer chromatography (HPTLC) fingerprint profile of Rasapachaka Yoga and Katutumbi Taila for their standardisation. Methods: Rasapachaka Yoga tablets and Katutumbi Taila were prepared according to classical methods described in Ayurveda texts and pharmacopeial standards. Organoleptic evaluation and physicochemical analysis were carried out following the guidelines of the Ayurvedic Pharmacopoeia of India (API) and Indian Pharmacopoeia (IP). HPTLC fingerprint profiling was performed using silica gel 60 F254 plates with Toluene: Ethyl acetate: Formic acid (5:2:0.5 v/v/v) as the mobile phase, and visualisation was done at 254 nm and 366 nm. Results:The organoleptic and physicochemical parameters of Rasapachaka Yoga and Katutumbi Taila were found to be within acceptable pharmacopeial limits, indicating the quality and stability of the formulations. HPTLC analysis revealed multiple peaks with distinct Rf values at 254 nm and 366 nm, indicating the presence of diverse phytochemical constituents such as alkaloids, flavonoids, phenolic compounds, glycosides, and terpenoids derived from the individual herbal ingredients. Conclusion: The HPTLC fingerprint profile generated in this study provides a characteristic phytochemical pattern for Rasapachaka Yoga and Katutumbi Taila. These findings can serve as reference standards for authentication, quality control, and pharmaceutical standardisation of these classical Ayurveda formulations.
