Nagaradi Choorna based transdermal patch: its formulation, physicochemical evaluation, HPTLC fingerprinting and In-vitro drug release study

Sweta Mer, Amal Thomas.

Abstract
ackground: The occurrence of pain and swelling in ankle sprain results in the tearing of ligaments. Current treatment methods involve the application of pain-relieving medication; however, the medication is not provided continuously. Nagaradi Choorna herbal preparation is widely used in Ayurveda medicine to treat inflammatory disorders. Objective: The present work aims at developing and evaluating the transdermal patch of Nagaradi Choorna along with the physicochemical studies, HPTLC fingerprint profiling, and in-vitro drug release studies of the formulated patches. Methods: The hydroethanolic extract of Nagaradi Choorna was prepared using ethanol and distilled water in 70:30 proportions with the drug-to-solvent ratio being 1:5. The transdermal patch of Nagaradi Choorna was prepared using the solvent casting technique wherein polyvinyl alcohol, polyethylene glycol-400, and dimethyl sulfoxide were used as the film-forming polymer, plasticiser, and permeation enhancer respectively. The physicochemical properties such as appearance, flexibility, homogeneity, and adhesion were determined for the formulated patches. Further, HPTLC fingerprint analysis was done to analyse the chemical constituents of the formula. Results: The prepared transdermal patch possessed good flexibility, adhesive strength, and homogeneity. Presence of phytoconstituents was found by HPTLC studies, and the formulation released drugs for a period of 72 hours. Conclusion: Transdermal patch formulation containing Nagaradi Choorna was developed by solvent casting technique. Satisfactory physical and chemical attributes and sustained release of the drug were observed from the formulated transdermal patch.
